기술개요

The present invention relates to the discovery of a novel miRNA that regulates a specific gene expressed during the chondrogenesis of human mesenchymal stem cells, and to the mechanism of action of the miRNA and the use of the miRNA in the preparation of an efficient cell therapy composition. In the present invention, a study was conducted to identify an miRNA that inhibits the gene Sox 9 expressed during the chondrogenesis of MSCs, and as a result, it was found that a specific miRNA, named “miR-XXX”, inhibits the expression of Sox 9, thereby inhibiting the chondrogenesis of MSCs. This invention can be utilized for the prevention and treatment of cartilage-related disease including degenerative arthritis.

Mesenchymal stem cells (MSCs) were treated with miR-XXX to induce chondrogenesis, and at 10 days after the start of induction of chondrogenesis, the glycosaminoglycan (GAG) content was measured, and the micromass was sectioned in paraffin and stained with safranin O. As a result, it was shown that miR-XXX inhibited the chondrogenesis of mesenchymal stem cells.

활용분야

This invention can be applied as stem cell therapy adjuvant for various bond damage diseases. And it can be used in cartilage regeneration of particular osteoarthritis, which has only a few practical therapeutic agents. The development of induction of differentiation of the cells into specific tissue stem cell can be used to new drug screening and establishing the system of drug toxicity analysis, and it will contribute to the basis of industrial application through finding gene and material for the differentiation and suppressor materials.
